  2023-10-24

F901318 is an orotomide, a novel class of antifungals, which inhibits pyridine biosynthesis by blocking dihydroorotate dehydrogenase (URA1) activity [137]. Although this pathway is conserved in humans, human URA1 is only 20% identical to its fungal homolog and it is inhibited 2000-fold less effectiv

Conclusion Acknowledgments and disclosure We thank Ms. Crystal Wylie for her help in the preparation of the manuscript. The development of the database upon which analyses of this study were dependent was supported by PO1 DC 00161 from the National Institute on Deafness and Other Communication

  2023-10-24

In this study, we have generated new FRET reporters to screen all submembrane domains of AT1R for interactions with CaM. We identified a new CaM-binding domain on the receptor’s SMD2 (a.a. 125 – 141), confirmed interactions at SMD4JM (a.a. 309 – 327) and characterized the full interacting domain in

  2023-10-24

This unsuspected antagonistic interaction of androgen with OXER1 could therefore explain the effect of plant derived compounds with this ligand-receptor system. For example, wedelolactone, a coumestan found in Eclipta alba (false daisy) and in Wedelia calendulacea that is the major component of ecli

The macrophage is the major cell type responsible for cholesterol uptake and metabolism associated with the initiation and progression of atherosclerosis in focal areas of the arterial subendothelial space. In advanced atherosclerotic lesions of cardiovascular disease, cholesterol-induced macrophage

  2023-10-24

APPL1 was the first adaptor protein identified that interacts directly with adiponectin receptors [18]. A two-hybrid study by Dong and colleagues revealed that the C-terminal extracellular domain of AdipoR1 interacts with adiponectin, whereas the N-terminal cytoplasmic domain of AdipoR1 interacts wi

  2023-10-24

Pituitary adenylate cyclase-activating polypeptide (PACAP) is a member of the secretin/growth hormone-releasing hormone (GHRH)/vasoactive intestinal peptide (VIP) family, with potent anti-inflammatory and potent cytoprotective properties [[6], [7], [8], [9], [10], [11], [12], [13]]. PACAP exists as

  2023-10-24

Introduction Adenosine deaminase (ADA), also known as adenosine aminohydrolase, is a key enzyme involved in the purine metabolism that converts adenosine to inosine irreversibly (Lupidi et al., 1997). It is a zinc containing metalloenzyme present in both prokaryotes and eukaryotes. In humans ADA

  2023-10-24

A key step in the ADAR reaction is the formation of the adenosine covalent hydrate with a tetrahedral center at C6. The nucleoside analog 8-azanebularine (box in Fig. 6), with its relatively high propensity to form a covalent hydrate, is useful as an adenosine replacement in ADAR2 substrates to trap

  2023-10-23

Our studies also reveal the normal functional roles of the MIR. Interaction between the MIR loop and the N-terminal α helix nucleates conformational maturation of AChR subunits, thereby promoting assembly of mature AChRs [52], [55]. This interaction in homologous parts of all AChR subunits appears t

• The subdivision of HT receptors

  2023-10-23

The subdivision of 5-HT receptors started in the 1950s by Gaddum and colleagues, when they realised that in the guinea pig ileum, the effects of 5-HT could be blocked in part by morphine (M), and in part by dibenzyline (D). Gaddum and Picarelli proposed two receptor classes, 5-HT M and 5-HT D (1957)

  2023-10-23

Skeletal pain associated with bone fracture is another area of critical need for a novel pain medication. NSAIDs and opiates are the two major therapies currently used to manage bone fracture pain. However, both of these therapies lead to significant unwanted side effects. Using the osteotomy model

  2023-10-23

ACL Regulation and Role in Lipid Metabolism ACL is a ubiquitous enzyme at the nexus of nutrient catabolism and synthesis of cholesterol and fatty acids. In mammals, it is highly expressed in lipogenic tissues including adipose, liver, and lactating mammary glands [9]. In the presence of ATP and C

  2023-10-23

Several studies have demonstrated that depletion of functional ATR increases the sensitivity of cancer cells to oncogene-induced replication stress thereby hindering tumour growth and inducing extensive cell death (Gilad et al., 2010, Murga et al., 2011, Schoppy et al., 2012). Importantly, Schoppy e
